AbstractA method was established for the determination of docetaxel (DOC) in human serum albumin (HSA) nanoparticles by the addition of cysteine. The effect of cysteine (the disulphide bond breaker) on HSA disulphide bonds was monitored by the detection on the content of DOC from HSA-NP using HPLC. The measurements were performed on addition of 1 mu g/L free cysteine to the samples, the content assay of DOC were carried out after an incubation period of 30 min at 37 degrees C under continuous shaking. The results showed that the addition of cysteine significantly increased the release of DOC front HSA-NP, and the determination results of DOC were 10% higher than that of acetonitrile.
